Abstract Over the last decade many new products containing metal nanoparticles have been introduced on the marked due to the properties of nano-sized particles. CuO nanoparticles (CuO NP’s) are amongst these metals and from their respective products some are eventually released into the aquatic environment. This can possibly be harmful to organisms living there. Though copper is an essential part of numerous important enzymes involved in vital biological processes, it can also lead to oxidative stress and cell death. If an organism experiences overexposure of the metal over a longer period of time, its body will try to cope by adjusting a broad arsenal of metal and ROS sensitive genes. In this project we investigate the expression of two such genes, Metallothioneine (MT) and Cytochrome C Oxidase (CCO). We used the freshwater oligochaete test-organism Lumbriculus variegatus to identify the possible, but yet undetected link between up or down regulation of the two genes and the overexposure to Copper. We used Cu2+ particles and CuO NP’s for two periods of time, 4 and 18 days to see if size and time had an impact. We found, that gene expression of CCO and metal contamination did not seem to be correlated. We did not achieve data for MT. We also succeeded in determining the subspecies of Lumbriculus variegatus due to a specific primer match with a subspecies 1 specific primer pair for COI-I (CCO).
