Vibrational Spectra of N-Butyryl-Homoserine Lactone

A special class of organic compounds, N-acyl homoserine lactones (HSL), is synthesized in bacteria and takes part in the intercellular signaling designated quorum sensing. The outcome of the intercellular signaling is responsible for many of the interesting properties which characterize colony forming bacteria such as adherence on surfaces, production of slime and resistance to a wide range of antibiotics. Biofilm formation on surfaces and medical devices poses a threat against public health. For instance, catheter related infections initiated by biofilms are common, costly and responsible for a substantial part of morbidity and mortality in many medical specialties. Lactones are also interesting compounds from a spectroscopic point of view. The spectroscopic information about these compounds in the literature is very sparse. In this study we present the Mid-infrared spectra of homoserine lactones in the solid state, and in liquid CCl4 solution. Quantum chemical calculations are performed in order to support the assignment of the measured vibrational bands and explain intermolecular interactions and solvent effects. The resulting designation of the reported spectral bands of C4-HSL lactone demonstrates the strength of computational chemistry as a tool for the study of complex mixtures of chemical substances. Composite spectra representing micro-crystalline aggregates and dissolved C4-HSL were measured and modeled. Based on the monomer structure form and cluster models of C4-HSL, computation of the combined spectra that resembles the measured one was possible. Here we report for the first time the key vibrational band of the monomer form.
