Optimal Temperature and pH of the Enzyme Beta-Glucosidase

The purpose of this report is to convey the findings of the investigation of the optimal conditions for the activity of the enzyme β-glucosidase from Aspergillus fumigatus at different temperatures and pH conditions by conducting enzyme assays. The temperature assay tested the enzyme with the substrate p-Nitrophenyl β-D- glucopyranoside and 50mM pH 5 sodium acetate buffer. The products of the reaction between this substrate and β-glucosidase are para-nitrophenol and glucose. Para-nitrophenol is yellow in colour under alkaline conditions. The pH assay tested the enzyme with the same substrate, but with a different buffers which were citrate-phosphate buffers at different pHs. The activity of the enzyme was determined by measuring the absorbance of the yellow product using a spectrophotometer. A calibration curve of para-nitrophenol was created, as well as graphs of the activity of β- glucosidase at different pHs and temperatures. Finally, the results of the experiments were then compared to literature results. The determination of these optima values can be used towards making the industrial conversion of cellobiose to biofuels more efficient.
